Researchers in the Faculty of Biology and Biotechnology (Maxim Shkurnikov, Stepan Nersisyan, Alexei Galatenko, Alexander Tonevitsky), along with colleagues from the Piragov Russian National Research and Medical University and Filatov City Clinical Hospital (Tatjana Jankevic, Ivan Gordeev, Valley Vechorko) The interconnect was studied between the HLA-I genotype and the severity of COVID-19.

Using machine learning, we built a model that provides an integrated assessment of the possible forces of the T cell immune response to COVID-19. Those individuals received a low risk score when the set of HLA-I alleles allowed effective presentation of SARS-CoV-2 viral peptides, whereas those with low presentation ability received a high risk score. (Range from 0 to 100). To validate the model, the genotypes of more than 100 patients and more than 400 healthy people (control group) suffering from COVID-19 were analyzed. The modeled risk score proved to be very effective in predicting the severity of COVID-19.

In addition to analyzing Moscow’s population, researchers used their model in a sample of patients from Madrid, Spain. This independent sample also confirmed the high accuracy of the prediction. Patients suffering from severe COVID-19 had a significantly higher risk score than patients with moderate to mild illness.

In addition to the correlations found between genotype and COVID-19 severity, the proposed approach assesses how specific COVID-19 mutations affect the development of T cell immunity to the virus.
